2Function of RecA
- Post replication repair
- Repairs dimers formed during replication or by
exposure to UV-light or chemicals - Part of the SOS response system in E. coli and
other organisms - Works with many other genes to repair mutations
3The SOS Response
- RecA is part of the SOS response, a repair
system. - SOS response repairs mismatched strands of DNA.
- RecA triggers this response by some mechanism
still unknown, but can do so in vitro in the
presence of ATP and single stranded DNA. - The response can be induced by UV crosslinking,
drugs, thymine deprivation, or mutations of dna
genes.
4The SOS Response
- Response includes activation of long patch
excision and Rec recombination repair pathways. - uvr genes and recA genes function together.
- Repair systems work with and stimulate nuclease
and polymerase systems.
5Why DNA repair is important
- Double mutant uvr- and recA- cells can only
tolerate about 2 dimers vs. 50 or more in wild
type cells. - Cell-cycle interference causing cessation of
mitosis or cancer.
6Symptoms of deficient DNA repair systems
- Some breast cancers caused by BRCA1 and uvr
- Xeroderma pigmentosum
- Ataxia-Telangiectasia
- Bloom Syndrome
- Cockayne Syndrome
- Fanconi Anemia

8How RecA Works
- LexA and recA are normally present at low levels
in the cell. The lexA product inhibits lexA,
recA, and many repair operons involved in the SOS
response. - When recA is stimulated, it causes autocatalytic
cleavage of lexA. - The genes involved in repair are now expressed as
well as increased levels of lexA and recA. - Some repair genes with 2 promoters express at
higher levels when the lexA inhibitor is
released.
9How RecA Works
- The SOS box is the target of usual lexA binding
and is found before all genes expressed in the
SOS response. These include - the expression of uvrA, B, C and D (dimer
excision repair genes) - recA is also responsible for homology searching
and recombination repair - ssb (protection of single stranded DNA)
- stiA (suppresses cell division)
- umuC and D (interact with DNA polymerase III to
allow error prone replication past damaged DNA)
and lexA is increased. - Increase expression of lexA and recA.

- lexA cleavage increases the concentration of recA
and lexA by about 50 fold. - This ensures that the response to repair by recA
is thorough and quickly halted by lexA when recA
is no longer stimulated.

- Bioinformatic analysis of recA and comparison
with other DNA repair/modifying proteins uvr,
rad, topoisomerases, and other radiation
activated proteins.

proteins. - The best conserved of these motifs is a
glycine-rich region, which typically forms a

alpha-helix. This loop interacts with one of the

31Conclusion
- RecA has a high degree of homology to RA_51 and
RA_23 which indicates that these proteins are
likely eukaryotic homlogs of the original RecA
repair protein. They show structural similarity
in their profiles, sequences, motifs, and
