InVitroCare SpermCare One-Step IUI System
  • InVitroCare SpermCare One-Step is a ready to use
    sperm preparation system for Intrauterine
    Insemination (IUI). The system includes
    pre-measured gradient in 15mL centrifuge tubes
    and pre-measured InVitroCare Sperm Wash Medium in
    glass vials.
  • The gradient, consisting of silane coated
    colloidal silica particles suspended in
    HEPES-buffered HTF, removes bacteria, epithelial
    cells, cell debris, immature sperm, and
    lymphocytes.
  • All system components contain glucose as a
    useable energy substrate to maintain normal cell
    metabolism and sperm function.

  • Bring all components of the system and semen
    sample to room temperature before use.
  • Step 1 ADD SEMEN
  • Gently layer 1-2 mL semen (at room temperature)
    on top of pre-measured SpermCare solution in blue
    cap 15 mL centrifuge tube.

    g) for 30 minutes.

  • Step 3 WASH
  • 3.1 Remove liquid layer, being careful not to
    disturb sperm pellet in bottom of centrifuge
    tube.
  • 3.2 Re-suspend sperm pellet by transferring the
    contents of green cap vial (InVitroCare Sperm
    Wash Medium) into centrifuge tube.
  • 3.3 Gently agitate.
  • 3.4 Centrifuge at approximately 2000 rpm (300-400
    x g) for 5 to 10 minutes.

  • Step 4 LOAD CATHETER
  • 4.1 Remove liquid layer, being careful not to
    disturb sperm pellet in bottom of centrifuge
    tube.
  • 4.2 Re-suspend sperm pellet by transferring the
    contents of silver cap vial (InVitroCare Sperm
    Wash Medium) into centrifuge tube.
  • 4.3 Gently agitate.
  • 4.4 Load entire volume (about 0.5-0.75 mL) of
    re-suspended sperm sample into an approved IUI
    catheter for subsequent transfer of the sperm to
    the uterine cavity.

  • Precautions and Warnings
  • Store reagents at 2-8C when not in use. Open and
    close bottles under aseptic conditions. Contents
    cannot be re-sterilized after opening. Centrifuge
    tube and vials are designed for single use.
  • All human, organic material should be considered
    potentially infectious. Handle all specimens as
    if capable of transmitting HIV or Hepatitis.
    Always wear protective clothing when handling
    specimens.
