  • As the development of technology, there are more
    and more methods in the biological field.
    However, these methods, such as absolute
    quantification and relative quantification, can
    confuse scientists and students. Here I will
    introduce the difference between the two methods.

  • Absolute quantification
  • The purpose of absolute quantification is to
    detect the number of molecular in the sample of
    aimed genes, which is called copy number. However
    the aim of the relative quantification is to
    detect the relative proportion of aimed genes in
    two or more samples. And there is no need to know
    the copy number in every sample.
  • Differences in experiement
  • In the experiemnet of the absolute
    quantification, it has to need the standard
    sample with the known copy number and the
    standard curve, which is not a big deal in the
    relative quantification experiement. There are
    two methods in relative quantification---standard
    curve and the aCT value comparation. If we use
    the first method, firstly, we should find out the
    absolute standard samples or the relative
    standard samples, which is much more convenient
    and it is easily to be operated. Whats more,
    relative quantification samples just need the
    proportion of DNA and RNA in samples without
    knowing the molecular number of standard samples.
    There is a typical method is to make the the

    method is a little complicated.

    soecificity and great repeatability. However, it
    is just suitable one determined sample and it is
    not cheap. And the absolute quantificationis
    always applied to determin the genes copy number,
    which is used in clinical diagnostic, such as,
    virus detection and nosophyte detection.
