Enzyme Introduction
  • 5,000 years ago, people stored milk in animal
    stomachs, in which milk can be turned into
    cheese. Enzymes have been used in the diagnosis
    of disease since the 1940's, derived from the
    intensive research in biochemistry. What are
    enzymes? How they work in biological reactions
    and living organisms? What are the applications
    of enzymes in biotechnology and medical research?

Enzyme Families
  • Enzymes are known to catalyze more than 5,000
    biochemical reaction types. Most enzymes are
    proteins, although a few are catalytic RNA
    molecules.
  • Enzymes are principally classified and named
    according to the reaction they catalyse. The
    chemical reaction catalysed is the specific
    property that distinguishes one enzyme from
    another, and it is logical to use it as the basis
    for the classification and naming of enzymes. The
    Nomenclature Committee of the International Union
    of Biochemistry and Molecular Biology (NC-IUBMB)
    classifies enzymes into families, using a four
    number code, on the basis of the reactions they
    catalyse.

Enzyme Assays
  • Enzyme assays are laboratory methods for
    measuring enzymatic activity, developing analysis
    technologies and studying enzyme catalytic
    mechanisms. They are vital for the study of
    enzyme kinetics and enzyme inhibition.
  • The enzyme activity is generally determined as
    substrate converted respectively product formed
    per time unit. Procedures for enzyme assays are
    documented or cited in various standard books and
    databases. However, the enzyme activity depends
    on manifold factors and general understanding of
    the particular features of enzymes is required,
    which cannot be described in all details in
    protocols for special enzyme assays. The common
    methods to be applied in enzyme assays are the
    subject of this section.
