Identification and Characterization of
Monoclonal Aptamer

• Selection of Polyclonal Aptamers
• Produce 5 pools from phases 1 with indexed
  primers for NGS sequencing
• NGS sequencing (105 to 106 reads per round) and
  identifying up to 20 sequences.

• Optimization of the Best Aptamer
• Truncate the best aptamer (1 aptamer) and test
  the binding properties of the variants.
• Provide suggestions for chemical modification(s)
  of the best variant, such as modification of the
  backbone to improve the nuclease resistance
  conjugation to a fluorophore

1. Synthesize and purify the top 5 monoclonal
   aptamer oligo candidate(s)
2. Quantitatively test each candidate for Kd
   affinities by SPR, BLI, ELISA, Microscopy, Flow
   Cytometry, etc.

• Library Enrichment (SELEX)
• Chemically synthesize and modify single-stranded
  DNA library or RNA library
• Iterative rounds of target screening (positive
  and negative), including small organic, peptide,
  protein, cell, and bacteria.

• Delivery and Report
• Supply 5 nanomoles of the best monoclonal
  aptamer or modified variant.
• Deliver project report containing all sequences
  from NGS sequencing.
