Title: Answers to Gel Electrophoresis Lab

1
Answers to Gel Electrophoresis Lab
  1. The colors green and black do not exist on the
gel after electrophoresis. They dont because
the green was broken down into blue and yellow.
The black was broken down into all colors (black
is a mixture of all colors)
  2. Yellow dye is the smallest molecule because it
was farthest away from the wells. It was able to
move the fastest and get farthest away

2
Answers contd
  • 5. Blue molecules were the biggest because they
    were closest to the wells
  • 6. The pigments have negative charge because
they move from the negative towards the positive
    end of the electrophoresis chamber.
  • 7. If used DNA would have to use restriction
enzymes to create the different sized pieces of
    DNA.

3
Answers contd.
  • 3 applications in which DNA fingerprint or gel
    electrophoresis may be useful
  • -- ID criminals
  • -- maternity/paternity
  • -- evolutionary relationships
  • -- ID remains of the dead
  • -- Army ID

4
For test
  • Dont need to know the exact results (for example
that blue dye was the biggest) need to know the
concepts (the bigger the molecule the less
    distance it moves)
  • Reread and study the procedure and background
    information for all labs

5
Answers to packet page 9-10
  • A, any cell would have same DNA
  • B. mRNA
  • C. Copy mRNA --gt ssDNA --gt dsDNA
  • D. DNA polymerase
  • E. Cut DNA (gene) and plasmid with same
    restriction enzyme

6
  • F, Cut plasmid and gene with R.E.
  • G. HIND III- palindrome exists at each end of
    gene and on plasmid
  • H. HIND III - must use same enzyme to cut
plasmid as was used to cut gene
  • I. They are the same
  • J. A ------- TTCGA
  • AGCTT A

7
  • K. The sticky ends are complementary
  • L. Make sure base pairs are complementary
  • M. Insert plasmid into bacterium (E. Coli)
  • N. The gene would either not have produced any
    protein or the incorrect protein

8
Are you my mother? Lab
  • Gel electrophoresis - used to determine
    variations in DNA sequences
  • The mom and and dads DNA fingerprint wont match
the babys exactly. This is because 1/2 the DNA
came from mother, 1/2 came from father

9
Review
  • 1. Be able to go from mRNA to DNA and DNA to
    mRNA
  • 2. What is ligase? Reverse transcriptase? DNA
    polymerase?
  • Whats reverse transcription?
  • What is a plasmid? Where can it be found? In
which of the labs was it needed?
  • What 2 things determine how a molecule (dye or
DNA) will move in gel electrophoresis?
