USE OF GENETIC DIVERSITY AND BIOTECHNOLOGICAL
ADVANCES IN FOREST TREE IMPROVEMENT
RESEARCH Project Director Dr. Ha Huy Thinh
12- RESEARCH RESULTS
- Breeding and trial establishment
- Data collection were done in progeny trials of P.
kesiya, Eucalypts, Acacias, Melaleucas. 20
families were collected to establish a
demonstration plantation in Lamdong. - Developed grafting techniques for 30 clones of P.
caribaea , P. massoniana and 30 clones of E.
urophylla - Study on flowering and control pollination
- Evaluated the flowering and seed production of P.
caribaea - Performed pollen collection and germination test
for P. merkusii and P. caribaea. Studied on
effect of moisture content and storage condition. - Controlled pollination between P. massoniana, P.
merkusii and P. caribaea, hybrid seed was
collected
13- Study on Biotechnology
- Creation of polyploidy, embryogenesis and study
on effect of physical aging and propagation
technique on the rooting - Using micro-satellite markers in evaluation of
genetic diversity and out cross-link rate in seed
orchards - Mating design and control pollination
- Published 5 papers in 2004

production of recombinant proteins for
agricultural and medical use Project Director
Dr Truong Nam Hai
18- Research results
- Part 1 Improving of enzyme activity of amylase
- Created a library of amylase mutant using
error-prone PCR and DNA shuffling. - Selected one amylase mutant with one amino acid
substituted and the amylase activity increased
twice compare to that of wild type.
- Part 2 Production of recombinant proteins of
Salmonella for vaccine and diagnostic - Cloned antigenic determinants of Salmonella as
fliC1/2/3 fljB1/2, gm1/2/3 and sef14 genes of S.
typhimurium and S. enteritidis - Expressed fljB, fljB2, gm1, gm2 genes encoding
for flagella antigen of S. typhimurium and S.
enteritidis in E. coli BL21.
fljB2
19- Part 3 Cloning and expression of Bacillus
collagenase - Constructed a genomic library of B. subtilis
FS-2 in E. coli DH5? - Selected one recombinant strain having
collagenase activity from a genomic library - Determined the sequence of cloned gene and its
restriction enzyme map - Cloning and expression of collagenase gene in E.
coli DH5?
Negative control pUC18
Collagenase activity of the selected recombinant
strain

25Comments
- We need the official appointment documents for
Subprograms Coordinator (Audit comment) - We need finance support from SIDA for projects
activity in time (Audit comment). - We have problem in VAT refund. We hope to solve
it in 2005.
