INTRODUCTION
CONCLUSION
OBJECTIVE
RESULTS
Clinical results In computer image analysis, the
formulation regimen containing 2 NAG plus 4 N
significantly reduced the appearance of facial
hyperpigmented spots vs. the vehicle regimen

N-acetyl glucosamine (NAG) 1 and niacinamide
(N) 2 have each been shown to reduce facial
hyperpigmentation in clinical evaluation. In
small base testing 1, a combination of the two
was observed to be more effective than
niacinamide alone. The current large base size
study of the combination was done to confirm
effectiveness of the combination.
A product regimen containing N-acetyl
glucosamine plus niacinamide clinically reduces
the appearance of facial hyperpigmented spots vs.
baseline and vs. the UV placebo
regimen. References 1. Bissett et al., AAD

METHODS
Double-blind, vehicle-controlled, full-face,
clinical study. Subjects with facial
hyperpigmentation (n 200, ages 40-60). March
May in Cincinnati, Ohio Vehicle regimen (n
100) AM moisturizer (SPF15) PM
moisturizer. Treatment regimen (n 100)
AM moisturizer containing 2 NAG 4 N
(SPF15). PM moisturizer containing 2 NAG
4 N.
All subjects used the same facial moisturizer
product in 2-week wash-out period and same facial
cleanser for entire study 2-week wash-out
period then 8-week (2X daily) treatment
period. Facial images captured at end
of the 2-week wash-out (baseline) after 4,
6, and 8 weeks of treatment. Hyperpigmented
spot image analysis of blind-coded images
Computer full color polarized image
analysis (hyperpigmented spot area) NCS
(non-contact full-face SIAscopeTM) for
hyperpigmented spot area and for
pigmentation evenness 3,4. No adverse events.
Additionally, significant reduction in spot
appearance by the 2 NAG 4 N formulation vs.
vehicle was noted in NCS NCS melanin evenness
(Figure 3). Figure 3 Evenness mean variance of
pigmentation brightness in NCS melanin map

Significant reduction in spot appearance by the
2 NAG 4 N formulation vs. vehicle was also
noted in NCS spot area fraction (Figure 2).
