Set Scale 1- open image in image J 2- zoom into
scale bar on image using zoom tool (right click)
3- draw linear region of interest (ROI) on scale
bar 4- go to gtanalyze gtset scale 5- type in
linear distance and units of scale bar, click
OK Measure object 6- zoom back out on image with
zoom tool (left click) draw another linear ROI
where you want to make a measurement 7- go to
gtanalyse gtmeasure to obtain measured distance
(this will be reported in pixels unless you have
set the scale as we just did)

When you make your measurement, remember that
our SEM stage tilts the sample so that the part
of the sample mapped at the bottom of the image
is closer to the gun than the part mapped at the
top of image (the stationary tilt axis is left
to right across image).

Use the terms depth of field and planar /
non-planar to describe why this sort of
measurement on an SEM image must be made and
reported with caution.
