Studies on the Hypoglycemic Constituents of
Liuwei Dihuang Pills and Nephrolepis multiflora
  • In this study, bioactive constituents of Liuwei
    Dihuang Pills and Nephrolepis multiflora (Roxb.)
    Jarrett ex Morton were investigated. n-BuOH
    extracts of Liuwei Dihuang Pills and CH2Cl2
    extracts of Nephrolepis multiflora (Roxb.)
    Jarrett ex Morton are obviously hypoglycemic
    activity. Rehmannia glutinosa Libosch. is the
    principal herbs (jun yao) of Liuwei Dihuang
    Pills. When Rehmannia glutinosa Libosch. (di
    huang) has been just prepared by drying and
    steaming. They are called sheng di huang (means
    raw) and shu di huang (means cooked),
    respectively. Investigating the changes in
    chemical components of Rehmannia glutinosa
    Libosch. during processing. Through all kinds of
    different solvents and chromatography system, a
    total of 22 compounds isolated. The known
    compounds which have isolated from the n-BuOH
    layer of Liuwei Dihuang Pills are
    5-hydroxymethyl-2-furaldehyde (1),
    2-hydroxy-5-hydroxymethylfuran (2),
    5-(a-D-Galactopyranosyloxy)methyl-2-
    furancarboxaldehyde (3), loganin (4),
    paeoniflorin (5), oxypaeoniflorin (6). The known
    compounds which have isolated from the n-BuOH
    layer of Rehmannia glutinosa Libosch. (sheng di
    huang) are catalpol (7), dihydrocatalpol (8),
    ajugol (9), scutelloside (10), uridine (11),
    sucrose (12), raffinose (13) and stachyose (14).
    Scutelloside (10) is the first report of
    Rehmannia glutinosa Libosch. As for the compounds
    which have isolated from the CH2Cl2 layer of
    Nephrolepis multiflora ( Roxb. ) Jarrett ex
    Morton are arctigenin (15), arctiin (16) and
    palmitic acid (17). In addition, the compounds
    which have isolated from the CH2Cl2 layer of
    Nephrolepis multiflora ( Roxb. ) Jarrett ex
    Morton are ethyl-ß-D-fructopyranoside (18),
    shikimic acid (19), ethyl shikimate (20),
    matairesinoside (21) and astragalin (22).
    Compound 1516 and 1921 are the first report of
    Nephrolepis multiflora ( Roxb. ) Jarrett ex
    Morton. Among them, 5-hydroxymethyl-2-furaldehyde
    (1), loganin (4), catalpol (7), dihydrocatalpol
    (8), raffinose (13), shikimic acid (19) and ethyl
    shikimate (20) can enhance glucose uptake in
    C2C12 experiment. Catalpol (7) is the indicator
    compound in sheng di huang. The content of
    dihydrocatalpol (8) in sheng di huang is more
    than the shu di huang. Therefore, the
    hypoglycemic activity of sheng di huang is better
    than the shu di huang.
