Abstract cSNAFL is a pH-sensitive fluorophore
routinely used in intracellular pH determination
at ambient pressures. With the goal of
characterizing cSNAFL for eventual high-pressure
studies on intracellular pH, properties of cSNAFL
under pressure are being studied for the first
time. The fluorescent spectrum of the dye cSNAFL
contains two emission peaks whose ratio varies
depending on the pH of the solution. The
emission spectrum of cSNAFL was recorded at
various pH and upon pressurization of an aqueous
buffer to 200 atm. Spectral peak ratios were
consistent with pressure-induced acidification,
indicating cSNAFL shows promise for use under
pressure.
