ERFP-Working Group Guidelines
Cryopreservation Final report
  • Sipke J. Hiemstra
  • Project co-ordinator
  • 4 September 2004, NC-workshop Bled

2
  • Objectives of project
  • Results
  • Evaluation of process and output
  • Financial report
  • Recommendations

3
Objectives (projectproposal november 2002)
  • Development of guidelines for cryopreservation of
    AnGR in Europe
  • Exchange of state of the art knowledge and
    experiences with cryopreservation
  • Common approach, adressing same type of questions
    and problems

4
Characteristics of guidelines cryopreservation
  • To support individuals or organisations to start
    or further develop cryopreservation programmes
  • Taking into account all major issues (eg legal,
    veterinary)
  • To support decision making regarding the design
    and operational choices
  • A practical guide, handbook, cookbook, checklist
  • Based on literature and experiences
  • European perspective
  • Not for law-making, but can create awareness
    among policymakers, stakeholders
  • Permanent draft

Final result 2004- Distributed April 2004-
Pdf available on internet

The process
  • December 2002 Decision of SC-ERFP to approve
    this project.
  • February 2003 Paris Workshop Cryopreservation
  • April 2003 first steps outline of
    guidelines-document
  • Two-day meeting of working group
  • 12/13 June 2003, Lelystad, the Netherlands
  • 13 participants from gt10 countries
  • Summer 2003 Synthesis of meeting and additional
contributions of working group members resulted
    in first draft
  • Progress report / presentation of first draft
guidelines at NC-workshop in Rome, 30 August
    2003
  • Sept 2003 March 2004 Chapter finalization and
    preparation of final report
  • Sept 2004 Final report at ERFP workshop Bled

Outline
  • Introduction
  • Fundamental considerations
  • Institutional setting and organisation
  • Intake of genetic material
  • Freezing and storage
  • Use of genetic material
  • Sanitary/veterinary requirements
  • Documentation
  • Legal issues

9
Fundamental considerations (2)
  • Conservation of genetic diversity on long term
    basis (CBD, ..)
  • Different perspectives, eg food security or
    cultural heritage
  • Balance between ex situ and in situ
  • Main objectives of cryopreservation (for future
    use)
  • Back up for genetic problems in living
    populations
  • Breed reconstruction
  • Creation of new lines or breeds
  • Back-up for future selection purposes
  • Research

10
Institutional setting and organisation (3)
  • Country specific strategies
  • Territorial organisation regional national
    European
  • Wide and active participation of stakeholders
  • Public and private responsibilities
  • Management
  • Funding
  • Cost benefit analysis
  • Limited information about cryopreservation in
    Europe

11
Intake (4)
  • The type and amount of the material to be stored
is a function of the specific objectives of the
cryobank, of the budgets that are available and
    of possible field constraints
  • Selection of species and breeds
  • Include existing collections
  • Many different criteria
  • Selection within breeds (depends on objectives!)
  • Sampling methods
  • Number of donor individuals
  • Type of genetic material
  • Amount of material
  • Time schedule

12
Freezing and storage (5)
  • Infrastructure (availability and choice)
  • Freezing protocols
  • Efficiency and efficacy
  • By species
  • By type of material
  • References

13
Use (6)
  • Property rights and rights of disposal
  • Operational criteria and procedures

14
Sanitary considerations (7)
  • How collection takes place
  • How storage is organised
  • Sanitary limitations for use
  • EU regulations by species and type of material
  • National regulations
  • Collected material with approved (inter)national
sanitary status versus collected material with
    no sanitary status

15
Documentation (8)
  • Vital for future use
  • Data on
  • Donor animal
  • Type of material
  • Veterinarian status
  • Legal status
  • Location of samples in storage
  • Software availabe
  • Linkage to other (in situ) databases

16
Legal issues (9)
  • Property rights and rights of disposal
  • Material Acquisition Agreements
  • Material Transfer Agreements
  • ? Legal instruments should be developed further
in order to guarantee long time viability of a
    cryopreservation program

17
Evaluation of the output
  • Comprehensive chapters with conclusive summary
  • At least useful checklist
  • All relevant issues together in one document
  • Differences between chapters
  • New issues
  • Based on extensive list of scientific research
  • Based on experiences
  • Valuable set of references by chapter
  • Document is not finished, (regular) improvement
    necessary
  • Feed back ?

18
Evalution of the process
  • Exchange of state of the art knowledge and
experiences resulted in a useful guide and can be
    distributed and discussed further
  • Network building in Europe and abroad
  • Exploration of new major issues
  • Valuable interaction of individuals with
    different background, expertise or experiences
  • Much can be done with some seed money, but
input of experts is limiting factor
  • Ownership of the output
  • Limited information about situation in all
    European countries
  • Follow up is recommended

19
Financial report
  • Budget 20.000 Euro
  • Expenses -/- 11.288 Euro
  • (4.921 Euro travel expenses) (3.256 Euro
    expenses Lelystad-meeting) (incl hotel, excl
    travel) (3.111 Euro expenses
    publication/distribution)
  • Balance 8.712 Euro

20
Conclusions and Recommendations
  • Working group made first attempt to assemble all
    relevant issues regarding cryopreservation
  • Useful guidelines but animal sector deserves
further development of some major issues
  • Co-operation between ERFP-network, EAAP-working
group and other expert/interest-groups is very
    fruitful
  • Last version available on ERFP website (pdf)
  • Collaboration with FAO and global network of NCs
  • More detailed information about situations in all
    European countries is necessary
  • Integration of ex situ and in situ strategies and
    programmes
