Sharing the KOMP Resources
  • Mutant Mouse Repositories

Clonal ES cell lines
NARRC January 2007
2
History of Mouse Resource Support
  • 1959 - NSF funded 1st JAX mouse repository
  • 1978 - Division of Research Resources
  • (pre-NCRR) funded the JAX mutant
  • mouse repository
  • 1999 - NCRR funded MMRRC to expand
  • mouse resource capability
  • 2006 - NIH funded
  • Repatriation of targeted mutation strains
  • Acquisition of Deltagen/Lexicon strains

NARRC January 2007

Value of KOMP Resources
  • Provide mutant mice and ES cells for
  • Basic research to basic clinician researchers
  • Understand pathways
  • Understand gene function
  • Identify drug targets
  • Phase 0 drug trials
  • Critical to archive and distribute knockout
    mouse resources

Partnerships for Sharing Resources
  • FIMRe - Federation of International Mouse
    Resources
  • Coordinate mouse resource archiving and sharing
  • Provide each other local distribution centers
  • Standardize protocols
  • Share advancements and training
  • IMSR - International Mouse Strain Resource
  • One-stop on-line shop for mouse resources
    available worldwide

Maintenance of resources
  • Living mice are not necessary - impossible except
    for strains in continuing demand
  • Gametes ES cells are mice in different formats
  • 2/3 of strains at JAX in frozen state only
  • 96 of strains in MMRRC in frozen state only
  • Both resource programs cryopreserve strains
    during the importation process
  • C57BL/6 sperm can now be reliably frozen and
    recovered by IVF
  • Frozen sperm enable recovery of large numbers of
    mice
