BacT/Alert Principle
  • LABS uses the BacT/Alert Blood Culture SystemTM
  • Samples are placed in sterile culture bottles
  • Bottle has a colorimetric sensor on the bottom to
    detect carbon dioxide as a measure of bacterial
    growth
  • If positive the sensor will change from
    blue/green to yellow

Media Used
  • BacT/Alert SATM (aerobic) Bottle
  • BacT/Alert SNTM (anaerobic) Bottle
  • or BacT/Alert PFTM (pediatric) Bottle
  • Store media at room temperature. Observe the
    expiration date. Do not use if expired.
  • Bottles from other manufacturers cannot be used
    on the BacT/Alert system. Cultures will be run
    manually.

Bottle Preparation
  • Inspect the bottle(s)
  • The broth should be clear (PF bottles contain
    charcoal)
  • The sensor (located in the bottom of the bottle)
    is blue-green or grey in color.
  • Do not use the bottle if the sensor is yellow or
    the broth is cloudy or if the bottle is cracked.
  • Do not use the bottles if past their expiration
    date
  • Remove the protective flip top overcap.
  • Cleanse the rubber stopper with 70 alcohol or
    iodine solution.
  • Allow to dry 1 minute before inoculation.

Collection
  • Follow Universal Precautions
  • Use good aseptic technique to avoid contamination
    of the sample
  • Proper skin or line disinfection is important

Specimen Volume
  • SA/SNTM bottles Fill with a maximum of 10 mL
    each
  • PFTM bottles Fill with a maximum of 4 mL
  • Filling with more that the maximum volume for
    each bottle may cause a false positive culture
  • Avoid injecting air into the bottles as this can
    affect the anaerobic conditions of the bottle,
    affecting test result

Label the bottles
  • Label the bottles with donor ID and an additional
    identifiers needed to positively match the
    samples with the requisition such as date, time
    and site
  • Do not cover the bottle barcodes or lot numbers
    with the labels

Ship or send the bottles to LABS
  • Do not refrigerate the bottles
  • Send to LABS as soon as possible

Reminders
  • Recommended blood to broth ratio is 15 to 110.
    As the blood volume increases, the yield of true
    positive cultures increases. Optimally, 20 mL of
    blood should be drawn from adults (10 mL per
    bottle).
  • Do not overfill the bottles, this may cause false
    positive readings.
  • For best volume control, mark the fill level on
    the side of the bottle prior to collection.
  • Use good aseptic technique to avoid contamination
    of the sample.
  • When labeling bottles, do not cover barcode
    labels or lot numbers.
  • Ensure that the bottles are positively labeled to
    match the requisition.
