The protein streptavidin (SA) binds strongly to
the small molecule biotin (vitamin H). Because
SA has four binding sites for biotin, it can be
used to as a linker between biotinylated
molecules in solution and biotins on a surface.
We have demonstrated patterning of SA on surfaces
using a pH-responsive polymer film and a
photoacid generator (PAG). Upon exposure of the
film (1) to deep UV light through a mask, the
polymer was chemically converted to reactive
aldehyde groups (1-2). Biotin was conjugated to
the surface (3), followed by SA (4) forming 18 x
18 mm feature sizes.
