• Paternity testing performed by Dept Haematology
  Cell biology since 1983.
• Initially only ABO Rh.
• Later other red cell groups (MN, Kell, Duffy,
  etc)
• HLA typing introduced 1984
• Red cell enzymes and serum proteins 1988
• Since 1999 only DNA analysis performed

OVERVIEW OF PATERNITY PROCEDURE

• EDTA sample collection (300 ul)
• DNA extraction
• DNA concentration determination
• DNA dilution (1 ul in 2000 ul)
• PCR set-up (1 ul of DNA dilution)
• Run PCR 4 h
• Denature and mix with internal std
• Electrophoresis on genetic analyzer 30
  min/sample
• Data analysis (duplicate) and comparison
• Report

CONCLUSIONS

• 400 450 trios tested each year
• Exclude about 40 of all men tested
• Exclude more men when male children are tested
• Probability of paternity always gt 99.99
