Step 1 Extract DNA from Banana
  • Follow the instructions listed on your handout.
  • (Steps 1-9)
  • Materials on the cart Amt you need
  • -Shampoo 1 teaspoon
  • -Salt 2 pinches
  • -Vial w/chilled 91 alcohol 1 vial/pair
  • -De-ionized water 20 ml
  • -Banana mixture 3 teaspoons
  • Important notes Reminders
  • -Note the appearance describe what you see.
  • -Rinse out your vials after you have made your
    observations
  • -Answer questions 1-4

  • Supplies are in the back of the room.
  • 1) Pick up a dixie cup w/ salt water (2 tsp)
  • 2) Swish around in your mouth thoroughly for 30
    sec.
  • 3) Spit back into the cup
  • 4) Pour the contents of the cup into the vial
    with soap solution.
  • 5) Gently rock the vial back forth (3 min)
  • 6) Take one pipette full of chilled alcohol and
    slowly add it to the vial while tilted making
    sure that they dont mix.
  • 7) Let stand for 1 minute
  • 8) Gently mix the alcohol into the soap layer
    with a glass rod.
  • 9) DNA should precipitate (whitish) Be patient

Final Steps
  • 10) Collect your DNA using a pipette or stirring
    rod and place into the capped containers. Add
    alcohol to preserve it.
  • 11) Seal and secure with glue.
  • 12) Write your initials on container (very small)
  • Place your DNA in the container w/your period
  • Pick up your DNA at lunch
  • 15) Clean up Rinse out the vials
  • Throw out paper cups
  • Please refill your vial with one pipette full
    of soap and place it back in the rack before you
    leave.
